The spread plate technique involves using a sterilized spreader with a smooth surface made of metal or glass to apply a small amount of bacteria suspended in a solution over a plate.
The spread-plate technique is analogous to the pour-plate procedure in its utility as a means to perform viable plate counts. However, because the colonies that form using the spread-plate technique are evenly distributed across the surface of the agar medium, cells from individual colonies can be isolated and used in subsequent experimental manipulations (e.g., as the inoculum for a streak-plate or a broth culture).
The most frequently used techniques are the CFU counting methods (spread-plate and pour-plate techniques, and the membrane filter technique) and the end point dilution method (MPN= Most Probably Number).
There are some advantages to using the spread plate method, one being that as colonies are formed on the surface of the agar, they are easier to sub-culture for confirmation tests. Also, as pre-poured solidified agar is used, there is no risk of exposure of the micro-organisms to heat stress, potentially caused by molten agar of too high a temperature.
