  • dark ground illumination - a method for the microscopic examination of living material, eg microorganisms, tissue culture cells, by scattered light. A special condenser with a circular stop illuminates the specimen with a numerical aperture larger than that collected by the objective. Only the scattered rays pass through the objective to reach the eye and the specimen appears luminous against a dark background.

Usage examples
  • Equal amounts of antigen and serum to be used in flat-bottomed microtitre plates, mixed and incubated at 30? C for two hours or 37? C for one to one and a half hours and the test read by low-power dark-field illumination using a magnification between 60 to 100.
